With over 700 putative ubiquitin E3 ligases, knowledge of the E3s capable of ubiquitylating your substrate is necessary information for a thorough understanding of protein biochemistry and also proper drug target identification. E3 ID™ employs a high-content microarray and LifeSensors' ubiquitin pathway expertise to identify candidate E3's that are capable of ubiquitylating your substrate. The service includes the following steps:

  1. A high-content, functional microarray is probed with client's substrate. Custom arrays are available upon request.
  2. A control array that has not been probed with the substrate is used.
  3. The experimental microarray is probed with the antibody to client's substrate or associated tag. Direct labeling of the protein is available for detection.
  4. The arrays are scanned and differences between the experimental array versus the control array are analyzed where locations of subtrate binding indicate E3-substrate interactions and potential E3 enzymes that ubiquitylate client's substrate. We report back a list of all proteins that show a statistically significant (p<0.05) difference between the two arrays. The protein list is also subjected to Gene Ontology analysis to sort by familial relationships. As we are not experts in the customer’s field, we leave it to them to filter the list(s) for proteins that may make biological sense.

The service can be performed with one of several commercially available arrays or a custom array.  LifeSensors can advise on the selection of the most appropriate array for the experiment. Screens have been performed successfully with protein arrays containing approximately 20,000 human proteins and over 495 E3's.

Array: Client-selected protein microarray

Client material required: Client's substrate (roughly 150 µL of a 100 nM solution), detection antibody for client's substrate or tag (optional). Direct labeling of the protein is available.
