Protease

Yeast is a cost-effective alternative for the expression of proteins that require an eukaryotic system to facilitate folding, yet it does not necessarily require advanced post-translational modification(s) associated with mammalian systems.

     The Saccharomyces Intracellular SUMOstar system allows for yeast strain selection with the Trp1 auxotrophic marker gene; it contains a high copy (2-micron) origin, and confers ampicillin resistance for selection in E. coli. Recombinant protein expression is driven by the tightly regulated, easily induced yeast metallothionein CUP1 promoter, which is easily regulated by copper. The use of this vector requires Saccharomyces strains that are trp1 and not copper sensitive.

     The methylotrophic yeast Pichia pastoris is known for its ability to produce very large quantities of recombinant protein, its propensity for high density growth in minimal media, post-translational modifications (e.g. glycosylation), and efficient protein secretion. However, production of recombinant proteins in P. pastoris is highly variable and greatly influenced by the inherent properties of the protein of interest. The Pichia Secretory SUMOstar system helps to eliminate this variability, and may greatly enhance protein production and purification. Expression is placed under the control of the strong, tightly regulated and methanol inducible alcohol oxidase promoter (AOX1).
